Recombinant prokaryotic lectin-Sia3 (RPL-Sia3) is the carbohydrate recognition domain (CRD) of rotavirus lectin expressed in, and purified from, E. coli. It is a monomer under physiological conditions and selectively binds to glycoproteins containing terminal ?-linked N-acetylneuraminic acid (Neu5Ac) groups. RPL-Sia3 activity requires metal ions, therefore, all buffers must be supplemented with 1 mM calcium chloride, magnesium chloride, and manganese chloride. It is recommended to use metal ion-supplemented Tris-buffered saline (TBS), pH 7.6, to dilute RPL-Sia3 to the desired working concentration (?20 µg/ml). Following dilution, incubate RPL-Sia3 for a minimum of 30 minutes prior to use to allow metal complexation.
