Recombinant prokaryotic lectin-Man2 (RPL-Man2) is a B. cenocepacia lectin expressed in and purified from E. coli.{65071} It is a monomer and selectively binds to glycoproteins containing terminal ?-mannose groups, such as glucose oxidase, over glycoproteins containing ?(1?6) fucose or ?(1?4) galactose groups. RPL-Man2 has been used to detect glycan structures in biolayer interferometry experiments. RPL-Man2 activity requires metal ions, therefore, all buffers must be supplemented with 1 mM calcium chloride, magnesium chloride, and manganese chloride. It is recommended to use metal ion-supplemented Tris-buffered saline (TBS), pH 7.6, to dilute RPL-Man2 to the desired working concentration (?20 µg/ml). Following dilution, incubate RPL-Man2 for a minimum of 30 minutes prior to use to allow metal complexation.
