Recombinant prokaryotic lectin-Fuc1 (RPL-Fuc1) is an A. fumigatus lectin expressed in and purified from E. coli.{65072,65281} It selectively binds to glycoproteins containing ?-linked fucose groups. RPL-Fuc1 has been used to detect glycan structures in microarrays and biolayer interferometry experiments. RPL-Fuc1 activity requires metal ions, therefore, all buffers must be supplemented with 1 mM calcium chloride, magnesium chloride, and manganese chloride. It is recommended to use metal ion-supplemented Tris-buffered saline (TBS), pH 7.6, to dilute RPL-Fuc1 to the desired working concentration (?20 µg/ml). Following dilution, incubate RPL-Fuc1 for a minimum of 30 minutes prior to use to allow metal complexation.
