DDD107498 is an antimalarial.{29310} It is an inhibitor of P. falciparum translation elongation factor 2 (PfeEF2) that inhibits protein synthesis. DDD107498 is active against P. falciparum strain 3D7 (EC50 = 1 nM) and clinical isolates of P. falciparum and P. vivax isolated from patients in areas where multidrug-resistant malaria is endemic (median EC50s = 0.81 and 0.51 nM, respectively). It reduces parasitemia in a mouse model of P. berghei infection (ED90 = 0.57 mg/kg), indicating activity against the blood-stage form of the parasite. DDD107498 is also active against liver schizont forms of P. berghei and P. yoelii (EC50 = ~1 nM) and eradicates P. berghei infection in mice when administered prior to infection at a dose of 3 mg/kg.
