Papain-like protease (PLpro) is a cysteine viral protease and component of the replicase complex in severe acute respiratory syndrome coronavirus (SARS-CoV) and SARS-CoV-2, the causative agents of SARS and COVID-19, respectively.{58386,58387} PLpro processes SARS-CoV and SARS-CoV-2 non-structural proteins 1-3, which, together with non-structural proteins 4-16, form the viral replicase complex, at three conserved sites, beginning with autolytic cleavage of itself.{57176,58386,58387} Covipyte™ EN450 is a fluorogenic substrate for PLpro that contains 9 amino acids, RELNGGAPI, which are recognized by PLpro. Upon enzymatic cleavage by PLpro, EDANS is separated from the Dabcyl quencher, displays excitation/emission maxima of 350/460 nm, respectively, and can be used to quantify PLpro activity.
