In the classical pathway of NF-?B activation, phosphorylation of the inhibitor of NF-?B (I?B?) releases the inhibitor from NF-?B, allowing I?B? degradation and NF-?B activation and nuclear import.{19822} BAY 11-7085 is an irreversible inhibitor of I?B? phosphorylation, preventing activation of NF-?B by cytokines and lipopolysaccharide (IC50 = 10 ?M).{16037} It blocks gene expression that is regulated through the classical pathway of NF-?B activation and in this way blocks apoptosis, cell adhesion, and inflammation.{16037,24580,24840} BAY 11-7085 is also used to study I?B? actions that are independent of NF-?B signaling.{24581}
