Amylin is a 37-residue peptide hormone secreted by pancreatic ?-cells that reduces food intake, modifies glycogen synthesis, slows gastric emptying, and increases satiety.{35231,38842,38841,38840} It binds to heteromeric complexes of human calcitonin receptor 2 (CTR2) with receptor activity-modifying protein (RAMP) 1 or 3 (IC50s = 7.86 and 6.35 nM, respectively).{39432} It also binds to mouse ?-thyroid-stimulating hormone thyrotroph (?-TSH) cells (IC50 = 10.5 nM) and rat nucleus accumbens membrane preparations (Ki = 0.024 nM).{38840,39432} In vitro, amylin (100 nM) inhibits insulin-induced, but not basal, glycogen synthesis in hepatocytes isolated from fasted rats.{38841} It also inhibits insulin-induced glycogen synthesis ex vivo in isolated soleus muscle from fasted rats.{38840} In vivo, amylin delays gastric emptying in rats (ED50 = 69 pmol) and decreases food intake in fasted mice (ED50 = 110 ?g/kg).{38842,38840} Unlike human amylin (Item No. 24274), rat amylin does not form fibrils and is not cytotoxic.{38843}
