Leukocyte-associated immunoglobulin-like receptor 2 (LAIR-2) is a soluble homolog of the membrane-bound inhibitory immune receptor LAIR-1.{56048} LAIR-2, which is comprised of a single immunoglobulin-like domain, binds to collagen and inhibits the LAIR-1/collagen interaction and subsequent activation of immunocytes.{56048,56047} It is produced by human peripheral blood mononuclear cells (PBMCs), monocytic and T cell lines, and activated CD4+ T cells.{56047,56050} LAIR-2 fused to the Fc region of human IgG1 inhibits collagen-induced platelet aggregation, adhesion of platelets to collagen-coated coverslips, and binding of collagen to von Willebrand factor (VWF) and glycoprotein VI (GpVI).{56049} Elevated levels of LAIR-2 have been found in the urine of pregnant women, the synovial fluid of patients with rheumatoid arthritis, and sera from patients with autoimmune thyroid diseases.{56047,56048} Cayman’s LAIR-2 (human, recombinant) protein can be used for binding assay applications. This protein consists of 131 amino acids, has a calculated molecular weight of 14.1 kDa, and a predicted N-terminus of Gln22 after signal peptide cleavage. By SDS-PAGE, under reducing conditions, the apparent molecular mass of the protein is 22 kDa due to glycosylation.
