Recombinant prokaryotic lectin-?Man (RPL-?Man) is a P. aeruginosa lectin expressed in and purified from E. coli. It binds to glycoproteins containing Lewis a (Lea), Lewis X (LeX), or terminal ?-mannose groups. RPL-?Man activity requires metal ions, therefore, all buffers must be supplemented with 1 mM calcium chloride, magnesium chloride, and manganese chloride. It is recommended to use metal ion-supplemented Tris-buffered saline (TBS), pH 7.6, to dilute RPL-?Man to the desired working concentration (?20 µg/ml). Following dilution, incubate RPL-?Man for a minimum of 30 minutes prior to use to allow metal complexation.
