Recombinant prokaryotic lectin-Gal1 (RPL-Gal1) is a lectin derived from the P. aeruginosa lectin PA-IL through mutagenesis, then expressed in and purified from E. coli.{65281} It is a dimer under physiological conditions and selectively binds to glycoproteins containing ?-linked galactose and N-acetyllactosamine (LacNAc) groups. RPL-Gal1 has been used to detect glycan structures in biolayer interferometry experiments. RPL-Gal1 activity requires metal ions, therefore, all buffers must be supplemented with 1 mM calcium chloride, magnesium chloride, and manganese chloride. It is recommended to use metal ion-supplemented Tris-buffered saline (TBS), pH 7.6, to dilute RPL-Gal1 to the desired working concentration (?20 µg/ml). Following dilution, incubate RPL-Gal1 for a minimum of 30 minutes prior to use to allow metal complexation.
