Recombinant prokaryotic lectin-?Gal (RPL-?Gal) is a P. aeruginosa lectin expressed in and purified from E. coli. It is a monomer and binds to glycoproteins containing terminal ?-linked galactose and/or N-acetylgalactosamine (GalNAc) groups. RPL-?Gal activity requires metal ions, therefore, all buffers must be supplemented with 1 mM calcium chloride, magnesium chloride, and manganese chloride. It is recommended to use metal ion-supplemented Tris-buffered saline (TBS), pH 7.6, to dilute RPL-?Gal to the desired working concentration (?20 µg/ml). Following dilution, incubate RPL-?Gal for a minimum of 30 minutes prior to use to allow metal complexation.
